Nucleotide sequence of a mouse tRNALeu gene
نویسندگان
چکیده
ORIGIN: The sequence was derived from i DBA-2 mouse genomic DNA fragment cloned in phage X£h4A (1) and was determined from overlapping M13 subdones as in Ref. 1. The sequence is the leftward end (as defined in Ref. 1) of a tRIAA gene cluster containing, in order, single genes encoding tRNA , tRNA A l p , tRNA G l y and tRNA l u , respectively. The (UNA, and t R N A * * coding regions are 338 bp apart Residues 430-480 correspond to residues 8-58 of the sequence reported in Ref. 1. Residues 1-7 of Ref. 1 were reported in error and are corrected here. The sequence shown below combined with that of Ref. 1 comprises 2221 contiguous bp of DNA. The tRNA gene is present in the opposite orientation relative to the other three genes of the cluster. Partial sequences of cognate rat gene clusters have been reported (2-5). FEATURES: The coding strand of the tRNA gene is shown, with the tRNA coding region in upper case (residues 137-219). Transcription is from right to left The terminal 3' CCA residues of the mature tRNA are not encoded. The anttcodon of the tRNA is 5"-CAG-3'. The tRNA coding region differs at one position in the extra arm from the rat sequence of Ref. 4 and one'position in the D-loop from the rat sequence of Ref. 3. The Hanking regions of the cognate mouse and rat genes are related, but quite variable in sequence. The t R N A ^ * gene is transcribed m the HeLa cell S-100 system in vitro.

Efficient Production of Biallelic RAG1 Knockout Mouse Embryonic Stem Cell Using CRISPR/Cas9
Background: Recombination Activating Genes (RAG) mutated embryonic stem cells are (ES) cells which are unable to perform V (D) J recombination. These cells can be used for generation of immunodeficient mouse. Creating biallelic mutations by CRISPR/Cas9 genome editing has emerged as a powerful technique to generate site-specific mutations in different sequences. Ob...

Genes and pseudogenes in a reiterated rat tRNA gene cluster.
A 13.4 kb rat genomic DNA fragment containing two related tRNA gene clusters was isolated from a rat lambda recombinant and analyzed for gene arrangement and nucleotide sequence. One cluster was found to contain a tRNALeuCUG gene while the second contained a tRNALeuCUA pseudogene with multiple base substitutions. The tRNALeu gene was found to possess an intact coding region and a functional tra...
متن کاملThe vlhA gene sequencing of Iranian Mycoplasma synoviae isolates
Mycoplasma synoviae expressed variable lipoprotein haemagglutinin (VlhA) is believed to play a major role in pathogenesis of the disease by mediating adherence and immune evasion. The aim of this study was sequencing Iranian M. synoviae isolates for the detection of nucleotide variation in the M. synoviae vlhA gene. Using oligonucleotide primers complementary to the single-copy conserved 5´ end...
متن کاملGene Expression under F8 Promoter Driving In Mouse Hepatoma Cells: A Step towards Gene Therapy of Hemophilia
Background and Objectives: Significant progress has been made in treatment of hemophilia. Ex-vivo gene therapy is going popular due to the capability of this method in using isogenic cells for genetic manipulation and reintroducing them into same host after proliferation. Most gene therapy techniques use viral vectors, which usually harbor a strong and non-specific promoter (e...
متن کاملSingle Nucleotide Polymorphism in the Dopamine Receptor Type 3 (DOP3) Candidate Gene Associated with Varroa Destructor Resistance in Honeybee
Extended Abstract Introduction and Objective: Varroa infestation is undoubtedly the greatest threat and challenge facing Apiculture today. This external parasite inevitably lives in the bee colony and causes irreparable damage to its colony and the subsequent honey production. One of the proposed strategies in this regard is the use of pesticides, which have a negative impact on the health of ...
